Italië, 1327. Het christendom is verdeeld door Paus Johannes XXII en keizer Louis IV van het heilige Romeinse Rijk. William van Baskerville komt samen met zijn volgeling Adso von Melk terecht in een afgelegen klooster in de Alpen. Daar worden ze geconfronteerd met een reeks mysterieuze moorden. Terwijl Baskerville en Melk de moorden onderzoeken, worden ze zelf opgejaagd door de genadeloze inquisiteur Bernard Gui, die diegenen vervolgd die kritiek hebben op de paus.
